Overview
This investigational combination therapy consists of hydroxychloroquine added to the standard-of-care regimen of peginterferon alfa-2a and ribavirin. Developed by Sheba Medical Center, it was evaluated in a Phase I/II clinical trial for patients with chronic hepatitis C genotype 1 who had previously failed to respond to standard dual therapy. Hydroxychloroquine, a small molecule antimalarial, was included for its potential to inhibit autophagy and modulate the immune response, theoretically enhancing the virological response. Peginterferon alfa-2a (Pegasys) provides antiviral and immunomodulatory effects by binding to type I interferon receptors, while ribavirin (Copegus) acts as a nucleoside analog that interferes with viral RNA replication through multiple mechanisms, including inhibition of viral RNA polymerase and depletion of intracellular guanosine triphosphate pools.
