A combination therapy consisting of hydroxychloroquine (HCQ) and temozolomide (TMZ). Temozolomide is an alkylating agent that causes DNA damage through methylation of adenine/guanine residues, while hydroxychloroquine functions as an autophagy inhibitor. The combination works by enhancing cell death in cancer cells, as blocking autophagy with hydroxychloroquine augments the cytotoxic effects of temozolomide. This approach has shown particular promise in treating advanced solid malignancies, especially metastatic melanoma and glioblastoma.
