Overview

Hydroxytyrosol + vitamin E is a combination of two antioxidant compounds—hydroxytyrosol, a phenolic compound primarily found in olive oil, and vitamin E (tocopherols/tocotrienols), a fat-soluble antioxidant. This combination has been studied for its potential therapeutic effects in non-alcoholic fatty liver disease (NAFLD), particularly for reducing liver fibrosis. Mechanistically, the combination acts by reducing oxidative stress and inflammation; it inhibits pro-fibrogenic gene expression in hepatic stellate cells activated by TGF-β signaling, reduces NOX2-mediated oxidative stress via interference with SMAD2/3 nuclear translocation/activation, and decreases circulating markers of fibrosis such as PIIINP. In both animal models and pediatric patients with NAFLD, supplementation with hydroxytyrosol plus vitamin E improved histological features of fibrosis as well as systemic markers of oxidative stress[1][2][6]. The combination is not an approved pharmaceutical drug but is being explored as a dietary supplement or nutraceutical intervention.
