HDCA + ME refers to a novel liposomal drug delivery system that encapsulates metformin within liposomes modified with hyodeoxycholic acid (HDCA). Metformin is a first-line biguanide used for type 2 diabetes that primarily acts by activating AMP-activated protein kinase (AMPK) to suppress hepatic glucose production. HDCA, a bile acid with a cholesterol-like structure, serves a dual role: it acts as a functional excipient to stabilize the liposomal membrane and as an active therapeutic agent that regulates glucose homeostasis, potentially through the activation of bile acid receptors like TGR5. This combination is designed to enhance the hypoglycemic efficacy of metformin while addressing its common limitations, such as poor gastrointestinal tolerance and low bioavailability. Preclinical studies have shown that these modified liposomes significantly improve glucose tolerance and protect liver tissue in diabetic mouse models.
