Drug intelligence / Profile preview
Iadademstat + atezolizumab is an investigational combination regimen consisting of iadademstat (ORY-1001), a small-molecule inhibitor of lysine-specific demethylase 1 (LSD1), and atezolizumab (Tecentriq), a programmed death-ligand 1 (PD-L1) blocking monoclonal antibody. Developed by Oryzon Genomics in collaboration with Genentech, this combination is being evaluated for the treatment of extensive-stage small cell lung cancer (ES-SCLC). The therapeutic rationale relies on iadademstat's epigenetic modulation, which increases the immunogenicity of SCLC cells by making them visible to the immune system and promoting the infiltration and activation of cytotoxic CD8+ T cells, thereby enhancing the anti-tumor efficacy of the immune checkpoint inhibitor atezolizumab. The combination is currently undergoing Phase I/II clinical evaluation, including studies sponsored by the National Cancer Institute (NCI) and investigator-initiated trials at Yale University.
