Drug intelligence / Profile preview
The combination of iadademstat, venetoclax, and azacitidine is an investigational three-drug regimen (triplet) being evaluated for the treatment of newly diagnosed, previously untreated acute myeloid leukemia (AML). This regimen combines iadademstat (ORY-1001), an orally active, highly selective small-molecule inhibitor of lysine-specific demethylase 1 (LSD1); venetoclax, a selective B-cell lymphoma-2 (BCL-2) inhibitor; and azacitidine, a hypomethylating agent. While the doublet of azacitidine and venetoclax is the standard of care for older or unfit patients with newly diagnosed AML, resistance and relapse remain common. The addition of iadademstat is designed to overcome resistance and enhance anti-leukemic efficacy by promoting differentiation and cell death in leukemia cells. The triplet is currently being evaluated in Phase I clinical trials, including the investigator-initiated ALICE-2 study sponsored by Oregon Health & Science University (OHSU) and a separate Phase I trial sponsored by the National Cancer Institute (NCI).
