iberdomide + daratumumab + dexamethasone

Iberdomide + daratumumab + dexamethasone is a combination therapy under clinical development for the treatment of relapsed or refractory multiple myeloma. Iberdomide is a novel, oral CELMoD (cereblon E3 ligase modulator) that induces ubiquitination and degradation of the transcription factors Aiolos and Ikaros, resulting in potent immunostimulatory and antimyeloma activity, greater than traditional IMiDs. Daratumumab is a monoclonal antibody targeting CD38, leading to immunemediated cell death and direct cytotoxicity of myeloma cells. Dexamethasone is a synthetic corticosteroid with cytotoxic and anti-inflammatory actions, widely used in multiple myeloma regimens to enhance the efficacy of combination partners and help reduce inflammation. This regimen has demonstrated manageable safety and promising efficacy in early-phase studies for patients with heavily pretreated or transplant-ineligible multiple myeloma. Current trials are evaluating its efficacy and optimal dosing compared to other combinations in multiple myeloma therapy[1][2][4][5][7][8].
