Overview

Iberdomide + dexamethasone is an investigational oral combination regimen used in the treatment of multiple myeloma, particularly relapsed or refractory cases. Iberdomide is a novel cereblon E3 ligase modulator (CELMoD) that has enhanced tumoricidal and immune-stimulatory effects compared to existing immunomodulatory drugs, while dexamethasone is a synthetic glucocorticoid corticosteroid used to suppress immune responses and inflammation. Iberdomide promotes ubiquitination and subsequent degradation of key transcription factors (Ikaros and Aiolos), enhancing anti-myeloma activity, immune cell activation, and apoptosis in malignant plasma cells. The combination is being studied in phase 1/2 trials and has demonstrated meaningful clinical activity, with overall response rates of about 26-32% in heavily pretreated patients with multiple myeloma who are refractory to multiple drug classes. Adverse effects include neutropenia, anemia, infection, and thrombocytopenia[1][5][7].
