Overview

Iberdomide + dexamethasone + carfilzomib is an investigational triple combination therapy studied for the treatment of relapsed or refractory multiple myeloma. Iberdomide is a novel oral cereblon E3 ligase modulator (CELMoD) that induces tumoricidal and immune-stimulatory effects. Carfilzomib is a second-generation proteasome inhibitor that blocks the chymotrypsin-like activity of the 20S proteasome, disrupting protein homeostasis and leading to apoptosis in myeloma cells. Dexamethasone is a synthetic corticosteroid with lympholytic and anti-inflammatory effects, widely used as part of multiple myeloma regimens. In preclinical models and phase 1/2 clinical trials, this combination (often referred to as IberKd) has shown manageable safety and promising anti-tumor activity in heavily pretreated relapsed/refractory multiple myeloma patients[1][3][5][7][9].
