Overview

**iberdomide + tafasitamab** is an investigational combination regimen for hematologic malignancies, specifically diffuse large B-cell lymphoma (DLBCL) and other B-cell non-Hodgkin lymphomas. *Iberdomide* (CC-220) is an oral small molecule immunomodulatory drug (IMiD) with enhanced activity compared to lenalidomide and pomalidomide. It works primarily through the binding and modulation of the cereblon E3 ubiquitin ligase, promoting the targeted degradation of transcription factors Ikaros and Aiolos, resulting in activation of T and NK cells, suppression of regulatory T cells, and increased anti-tumor immunity. *Tafasitamab* (MOR208, brand name Monjuvi) is an Fc-modified monoclonal antibody targeting CD19, a surface protein on B cells. Tafasitamab acts via direct cytotoxicity—inducing apoptosis and programmed cell death in B cells—and through immune-mediated mechanisms including antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP)[2][4][6]. The combination is being tested to enhance efficacy through complementary immunological mechanisms: iberdomide stimulates the immune system, while tafasitamab specifically targets B-cell malignancies for destruction by immune effector cells.
