IBI-322 + lenvatinib is an investigational combination therapy for cancer that pairs IBI-322—a recombinant humanized bispecific monoclonal antibody targeting both CD47 and PD-L1—with lenvatinib, a small molecule multi-targeted tyrosine kinase inhibitor. \nIBI-322 is designed to block the immune checkpoint pathways mediated by CD47/SIRPα and PD-1/PD-L1. By binding preferentially to tumor cells co-expressing CD47 and PD-L1, it stimulates macrophage-mediated phagocytosis (innate immunity) and enhances T-cell activation (adaptive immunity), while minimizing toxicity to normal cells such as red blood cells[3][4][5][6]. Lenvatinib inhibits multiple receptor tyrosine kinases involved in tumor angiogenesis and proliferation. The combination aims to provide synergistic antitumor effects by simultaneously modulating the immune microenvironment and inhibiting tumor growth signaling pathways.
