Overview
IBI322 + decitabine is an investigational combination therapy consisting of IBI322, a recombinant human bispecific monoclonal antibody targeting both CD47 and PD-L1, and decitabine, a small-molecule DNA methyltransferase inhibitor. IBI322 is designed to disrupt the “don’t eat me” signal (CD47/SIRPα axis) and block PD-L1-mediated immune evasion, thereby enhancing antitumor immune responses. Decitabine acts as an epigenetic modulator, inducing DNA hypomethylation and promoting tumor cell differentiation and apoptosis. The combination is primarily investigated for hematologic malignancies such as acute myeloid leukemia (AML), leveraging potential synergistic effects between immune checkpoint blockade and epigenetic therapy[4].
