Overview

IBI325 + sintilimab is a combination of two monoclonal antibodies developed for cancer immunotherapy. IBI325 is a humanized antibody targeting CD73, an ecto-enzyme that converts extracellular AMP to adenosine, which suppresses anti-tumor immune responses by activating adenosine A2A and A2B receptors on immune cells. By inhibiting CD73, IBI325 aims to relieve the immunosuppressive tumor microenvironment. Sintilimab is a fully human IgG4 monoclonal antibody against programmed cell death protein 1 (PD-1), blocking its interaction with ligands and restoring T-cell mediated anti-tumor immunity. The combination is being investigated for advanced solid tumors, leveraging dual blockade of immunosuppressive pathways to enhance anti-cancer efficacy[1][2][4][5].
