IBI343 + mFOLFOX is a combination therapy being developed by Innovent Biologics for the treatment of CLDN18.2-expressing solid tumors, including gastric, gastroesophageal junction, and pancreatic cancers. IBI343 is a recombinant human anti-CLDN18.2 monoclonal antibody-drug conjugate (ADC) utilizing a cleavable linker to attach an exatecan payload (a topoisomerase I inhibitor) with a drug-to-antibody ratio (DAR) of 4. The mFOLFOX component is a standard chemotherapy regimen consisting of oxaliplatin, leucovorin, and 5-fluorouracil. The combination aims to leverage the targeted delivery of IBI343 and its bystander killing effect alongside the systemic cytotoxic activity of mFOLFOX. It is currently being evaluated in Phase 1 clinical trials (NCT05458219).
