Overview

A combination regimen comprising IBI351 (also known as GFH925), cetuximab, and FOLFIRI (a multi-agent chemotherapy regimen consisting of folinic acid, fluorouracil, and irinotecan). - **IBI351** is a small molecule, orally active, covalent, and irreversible inhibitor of KRAS G12C, designed to target and modify the KRAS G12C protein and block downstream signaling, resulting in tumor cell apoptosis and cycle arrest. - **Cetuximab** is a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR), inhibiting its signaling and recruiting immune effector cells for the destruction of EGFR-expressing tumor cells. - **FOLFIRI** combines folinic acid (leucovorin, a vitamin B derivative that enhances fluorouracil), fluorouracil (a pyrimidine analog antimetabolite disrupting DNA synthesis), and irinotecan (a topoisomerase I inhibitor causing DNA damage). This regimen is being investigated for advanced/metastatic solid tumors, particularly metastatic colorectal cancer harboring KRAS G12C mutations, and leverages targeted therapy (KRAS G12C and EGFR) with cytotoxic chemotherapy[1][2][3][4][5][6][7][8].
