Overview

IBI363 + bevacizumab is a combination therapy under clinical investigation for advanced colorectal cancer and other solid tumors. The regimen combines IBI363, a first-in-class PD-1/IL-2α-bias bispecific antibody fusion protein, with bevacizumab, an anti-VEGF monoclonal antibody. IBI363 is designed to both block the PD-1/PD-L1 immune checkpoint pathway and activate the IL-2 signaling pathway by fusing an anti-programmed death receptor 1 (PD-1) antibody with interleukin-2 (IL-2). This dual mechanism aims to reverse T-cell exhaustion and promote activation of T cells and natural killer (NK) cells within the tumor microenvironment. Bevacizumab inhibits angiogenesis by targeting vascular endothelial growth factor A (VEGF-A), thereby restricting tumor blood supply. Clinical studies have shown that this combination demonstrates promising anti-tumor efficacy with durable responses in patients with advanced colorectal cancer—including those who are non-MSI-H/dMMR or have RAS mutations—and maintains a manageable safety profile[1][3][9]. The combination is being evaluated in Phase 1 trials for advanced malignancies including colorectal cancer[4], melanoma, and non-small cell lung cancer[2].
