Drug intelligence / Profile preview
IBIO-610 + semaglutide is an investigational combination therapy in preclinical development for the treatment of obesity and cardiometabolic diseases. Developed by iBio, the combination consists of IBIO-610, a potentially first-in-class monoclonal antibody targeting the Activin E ligand (encoded by the INHBE gene), and semaglutide, an approved glucagon-like peptide-1 (GLP-1) receptor agonist. IBIO-610 is designed to inhibit active Activin E to promote fat-selective weight loss and preserve lean muscle mass during GLP-1-driven weight loss. In preclinical studies in obese non-human primates, a single dose of IBIO-610 combined with twice-weekly semaglutide reduced active Activin E levels by approximately 97% and significantly improved the quality of weight loss, with fat accounting for 93% of the weight lost compared to 77% with semaglutide alone. Due to its extended half-life, IBIO-610 has the potential to support twice-yearly dosing in humans. First-in-human clinical trials are planned to begin in the first half of 2027.
