Overview

BTK inhibitor + Anti-CD19 CAR T cells is an investigational combination therapy regimen that pairs the small-molecule Bruton's tyrosine kinase (BTK) inhibitor ibrutinib with chimeric antigen receptor (CAR) T cells targeting the CD19 antigen. Ibrutinib, a covalent inhibitor of BTK, disrupts B-cell receptor signaling and has been shown to enhance CAR-T cell function by promoting proliferation, enriching for less-differentiated T-cell phenotypes, and downregulating exhaustion markers. Additionally, ibrutinib may reduce the severity of cytokine release syndrome (CRS), a common side effect of CAR-T therapy. This combination is primarily being evaluated for the treatment of relapsed or refractory B-cell malignancies, including B-cell non-Hodgkin lymphoma (B-NHL), where it aims to improve the overall response rate and durability of treatment compared to CAR-T therapy alone.
