Overview

**Ibrutinib + paclitaxel** is an investigational combination therapy consisting of ibrutinib, a selective irreversible inhibitor of Bruton's tyrosine kinase (BTK), and paclitaxel, a microtubule-targeting antineoplastic agent. Ibrutinib blocks B-cell receptor signaling through BTK inhibition, impacting cancer cell proliferation, survival, and tumor microenvironment modulation. Paclitaxel stabilizes microtubules, thereby inhibiting cell division and inducing apoptosis in rapidly dividing cells. Preclinical studies demonstrate potential synergy, as ibrutinib can sensitize tumor cells to paclitaxel by inhibiting drug efflux mediated by ABCB1 and ABCC10 transporters. The combination is under investigation for treatment of solid tumors, notably urothelial carcinoma and various resistant cancers, but is not an approved regimen[2][4].
