icaritin + pexastimogene devacirepvec

Icaritin + pexastimogene devacirepvec (Acoradine + Pexa-Vec) is an investigational combination therapy being co-developed by Beijing Shenogen Biomedical and Lee's Pharmaceutical for the treatment of advanced hepatocellular carcinoma (liver cancer). The combination regimen pairs icaritin (Acoradine), an orally active small molecule immunomodulator derived from the traditional Chinese medicine Epimedium that acts as an estrogen receptor alpha (ERα) antagonist and BHLHE40 inhibitor, with pexastimogene devacirepvec (Pexa-Vec), an oncolytic vaccinia virus engineered to selectively replicate in tumor cells and express human granulocyte-macrophage colony-stimulating factor (GM-CSF). Together, these agents exert synergistic anti-tumor activity by directly lysing cancer cells, disrupting tumor vasculature, and stimulating a robust systemic anti-tumor immune response. The combination is currently in Phase 2 clinical development in China.
