Icotinib + chemotherapy refers to the combination of the small molecule epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor icotinib with standard cytotoxic chemotherapeutic agents. Icotinib selectively inhibits EGFR tyrosine kinase activity, blocking downstream signaling pathways involved in tumor cell proliferation and survival. In clinical studies, this combination has been primarily investigated as a first-line treatment for advanced non-small cell lung cancer (NSCLC) with sensitive EGFR mutations. The most commonly studied chemotherapies in this context are pemetrexed and carboplatin or cisplatin and paclitaxel. The rationale is to enhance antitumor efficacy by targeting both EGFR-driven signaling and DNA replication/cell division mechanisms. Clinical trials have shown that combining icotinib with chemotherapy improves progression-free survival compared to monotherapy but may increase certain adverse events such as leukopenia and liver function damage; these side effects are generally manageable[1][3][5].
