This is a combination regimen consisting of three drugs—idarubicin, azacitidine, and venetoclax—used primarily in the treatment of acute myeloid leukemia (AML). Idarubicin is an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II, leading to DNA damage and cell death. Azacitidine is a hypomethylating agent that incorporates into DNA and RNA, inhibiting DNA methyltransferase and resulting in hypomethylation of DNA, which can restore normal gene function in cancer cells. Venetoclax is a selective B-cell lymphoma-2 (BCL-2) inhibitor; it promotes apoptosis by blocking the anti-apoptotic protein BCL-2. The combination aims to improve remission rates by targeting multiple pathways involved in leukemic cell survival and proliferation. This regimen has been studied for newly diagnosed AML patients who may not be candidates for intensive chemotherapy[1][5].
