Overview

IDE397 + paclitaxel is an investigational combination therapy comprising **IDE397**, a potent and selective small molecule inhibitor of methionine adenosyltransferase 2 alpha (MAT2A), and **paclitaxel**, a well-established microtubule-stabilizing chemotherapy. IDE397 selectively targets tumors with methylthioadenosine phosphorylase (MTAP) deletion, exploiting a metabolic vulnerability in these cancers. Paclitaxel disrupts cell division by stabilizing microtubules. This combination is being clinically investigated for the treatment of solid tumors, particularly **esophagogastric cancers**, with a mechanistic rationale based on complementary antitumor actions and the potential to enhance efficacy in MTAP-deleted tumors[1][4].
