idelalisib + everolimus

Idelalisib + everolimus is a combination of two orally administered small molecule inhibitors under investigation for hematological malignancies, particularly relapsed/refractory mantle cell lymphoma (MCL). Idelalisib is an inhibitor of the phosphatidylinositol 3-kinase delta (PI3Kδ) isoform, which is highly expressed in lymphocytes, where it disrupts critical survival and proliferation pathways for malignant B cells. Everolimus is an inhibitor of the mammalian target of rapamycin (mTOR), a kinase involved in cell cycle progression and angiogenesis, through binding to FKBP-12 and inhibition of downstream effectors. The combination is being evaluated primarily for its additive or synergistic antitumor activity and tolerability in relapsed/refractory B-cell malignancies[3][7].
