idronoxil + carboplatin

This is a **combination therapy** consisting of **idronoxil** (an isoflavone derivative, also known as phenoxodiol), formulated as a rectal suppository (NOX66), administered in combination with **carboplatin**, a platinum-based chemotherapeutic agent. Idronoxil acts primarily by **inhibiting ENOX2 (ecto-NOX disulfide-thiol exchanger 2)**, impairing tumor cell resistance and enhancing apoptosis, with some immune modulation effects by activating both innate and adaptive immunity. As a chemosensitizer, idronoxil has shown potential to reverse or reduce resistance to cytotoxic agents such as carboplatin, thereby increasing the efficacy of chemotherapy in refractory solid tumors. Carboplatin, meanwhile, is a DNA alkylating agent inducing inter- and intra-strand DNA crosslinks, triggering tumor cell death. The combination is under clinical investigation for refractory solid tumors and platinum-resistant ovarian cancer, with early results indicating a tolerable safety profile and disease stabilization in several cases. NOX66 is being developed and investigated primarily by Noxopharm.
