**IFN-γ mRNA@βGlus-LNPs** is an orally administered, β-glucan-functionalized lipid nanoparticle formulation carrying messenger RNA encoding interferon gamma. Developed in a preclinical triple-negative breast cancer mouse model, the particles use β-glucan-mediated uptake through Dectin-1-expressing intestinal M cells in Peyer's patches, after which endogenous macrophages transport the mRNA cargo through lymphatic and systemic circulation to tumors. Local translation of the cargo produces transient interferon gamma expression in the tumor microenvironment, promoting pro-inflammatory M1-like macrophage reprogramming and cytotoxic T-cell activation.
