Overview

IG-002 is a T-cell receptor mimic (TCRm) bispecific antibody designed to target the intracellular neoantigen KRAS G12V. It specifically recognizes the KRAS G12V mutated peptide when presented on the cell surface by the Human Leukocyte Antigen (HLA)-A*02:01 molecule. By mimicking the natural recognition mechanism of a T-cell receptor but with the high affinity and stability of an antibody, IG-002 bypasses the need for complex cell therapy manufacturing. The bispecific format includes a CD3-binding domain that recruits and activates cytotoxic T-cells directly to the tumor site, leading to the selective lysis of KRAS G12V-positive cancer cells. This approach is particularly relevant for solid tumors such as pancreatic, colorectal, and lung cancers where KRAS mutations are prevalent and difficult to target with conventional antibodies. IG-002 was developed by ImmunoGenesis based on technology originally discovered at Johns Hopkins University.
