The **Ii-Key-H1 Swine Influenza Vaccine (G4EA H1N1)** is an experimental epitope-optimized vaccine designed to elicit broad and enduring immunity against swine influenza A virus (IAV-S) subtype H1N1, specifically targeting the genotype 4 Eurasian avian-like (G4 EA H1N1) strain. It employs the **Ii-Key** technology, which enhances MHC class II presentation of peptide epitopes derived from hemagglutinin (HA) to promote robust CD4+ T cell responses, combined with an **epitope optimization approach (Epigraph)** that computationally designs immunogens incorporating the most prevalent T and B cell epitopes from diverse H1 IAV-S sequences spanning 1930–2021. Developed to address the high genetic diversity of H1 strains predominant in global swine populations, preclinical studies in pigs demonstrated superior cross-reactive T cell responses (IFN-γ), reduced clinical disease, lower viral shedding, and decreased lung lesions compared to wild-type or commercial vaccines. G4 EA H1N1 itself poses a pandemic threat due to efficient human receptor binding (α2,6 sialic acid), replication in human airway cells, aerosol transmission in ferrets, and antigenic drift from human vaccine strains.[1][2][4]
