IL-10-Fc + alpha-PD-1 is a preclinical combination therapy developed by Leman Biotech. It consists of an IL-10-Fc fusion protein (specifically Leman's Meta 10 technology) and an anti-PD-1 monoclonal antibody. The mechanism of action involves IL-10-mediated metabolic reprogramming of T cells, which enhances oxidative phosphorylation and prevents or reverses T-cell exhaustion, thereby synergizing with the checkpoint blockade provided by the anti-PD-1 component. In preclinical studies using the CT26 colorectal cancer mouse model, the combination demonstrated a 60% cure rate, suggesting potential for treating tumors that are typically resistant to checkpoint inhibitor monotherapy.
