Overview

IL-15-armored chimeric antigen receptor (CAR) T-cell therapy is an investigational adoptive cell therapy developed by The First Hospital of Jilin University for the treatment of relapsed or refractory multiple myeloma and plasma cell leukemia. This platform involves engineering T cells to express CARs targeting specific antigens associated with plasma cell malignancies—namely B-cell maturation antigen (BCMA), CD19, and GPRC5D—while also incorporating the genetic sequence for Interleukin-15 (IL-15). IL-15 is a critical cytokine for the survival and homeostatic proliferation of T cells and natural killer (NK) cells. By "armoring" the CAR-T cells with IL-15, the therapy aims to improve the persistence of the cells in the patient's body and enhance their ability to combat the immunosuppressive environment of the bone marrow. The clinical program evaluates single-target (BCMA or GPRC5D) and dual-target (CD19/BCMA) constructs to provide multiple therapeutic options for patients who have failed standard treatments.
