IL-15 DNA + bupivacaine is an investigational combination therapy that includes a DNA plasmid encoding interleukin-15 (IL-15) and the local anesthetic bupivacaine. IL-15 is a cytokine with potent immune-stimulating properties, particularly in activating natural killer (NK) cells and CD8+ T cells, making it a promising agent for cancer immunotherapy[6]. Bupivacaine is an amide-type local anesthetic that blocks voltage-gated sodium channels on neurons, preventing nerve impulse transmission and providing local anesthesia[3][5]. In this combination, bupivacaine may be used to enhance the delivery or tolerability of IL-15 DNA at the injection site. The primary indication under investigation for this combination is cancer immunotherapy; however, as of now, no IL-15 agonist drug or therapy has been approved for marketing[6].
