Overview

This is a combination therapy consisting of interleukin-2 (IL-2), an anti-tumor necrosis factor (anti-TNF) agent, and thalidomide. Each component has distinct immunomodulatory properties: - **Interleukin-2 (IL-2)** is a cytokine that stimulates the growth and activity of T cells, enhancing immune responses against tumors. - **Anti-TNF agents** are biologics or small molecules that inhibit tumor necrosis factor-alpha (TNF-alpha), a key pro-inflammatory cytokine involved in autoimmune diseases and cancer-related inflammation. - **Thalidomide** is an immunomodulatory drug with anti-inflammatory, antiangiogenic, and antineoplastic effects. It inhibits TNF-alpha production, modulates other cytokines such as IL-6, shifts T helper cell responses from Th1 to Th2 type, and provides costimulatory signals to T cells leading to increased IL-2 production at higher doses[5][7][8]. The combination aims to leverage the immune-stimulating effects of IL-2 with the anti-inflammatory/antiangiogenic actions of thalidomide and TNF inhibition for synergistic antitumor or immunoregulatory effects. This regimen has been studied primarily in oncology settings such as metastatic renal cell carcinoma[1][2], but also has theoretical applications in other inflammatory or neoplastic conditions.
