IL-21 modulated MART-1-specific autologous CD8+ T cells is an investigational adoptive cell therapy developed by the Fred Hutchinson Cancer Center for the treatment of metastatic melanoma. The therapy involves the isolation of a patient's own CD8+ T cells, which are then stimulated ex vivo with interleukin-21 (IL-21) to enhance their anti-tumor activity, persistence, and effector function. These T cells are specifically selected or engineered to recognize the MART-1 (Melanoma Antigen Recognized by T cells-1) antigen, a protein commonly expressed on the surface of melanoma cells. In clinical protocols, these therapeutic lymphocytes are typically administered intravenously following a lymphodepleting conditioning regimen of cyclophosphamide and are accompanied by systemic aldesleukin (IL-2) to support the expansion and survival of the infused cells in vivo.
