IL-24S + IL-15 is a novel recombinant chimeric fusion cytokine consisting of an engineered, highly secreted version of Interleukin-24 (IL-24S or Superkine) and Interleukin-15 (IL-15) connected by a flexible linker. Developed by researchers at Virginia Commonwealth University, this fusion protein is designed to provide a dual-action therapeutic effect against cancers such as glioblastoma. The IL-24S component induces cancer-specific apoptosis and toxic autophagy without affecting normal cells, while the IL-15 component promotes the activation, proliferation, and recruitment of natural killer (NK) cells and CD8+ T cells. In preclinical models, the fusion protein has been delivered via an adenoviral vector (Ad5) combined with focused ultrasound and microbubbles to penetrate the blood-brain barrier and target intracranial tumors.
