IL-7 + IL-15 + iMSCs (also known as ERNA-101) is a preclinical cell-based biologic therapy being developed by Ernexa Therapeutics. The therapy consists of induced pluripotent stem cell-derived mesenchymal stem cells (iMSCs) engineered to secrete the pro-inflammatory cytokines interleukin-7 (IL-7) and interleukin-15 (IL-15). These synthetic, allogeneic cells are designed to home to the tumor microenvironment (TME) and deliver high local concentrations of cytokines, thereby activating and expanding T cells and natural killer (NK) cells in situ. Additionally, the therapy promotes the polarization of immunosuppressive M2-like macrophages toward a pro-inflammatory M1-like phenotype, effectively converting 'cold' tumors into 'hot' ones. ERNA-101 is primarily being investigated for the treatment of advanced solid tumors, including platinum-resistant ovarian cancer and triple-negative breast cancer, with potential applications in hematological malignancies.
