IL2-Pt@Nanogel is an experimental chemoimmunotherapy nanogel designed for the dual delivery of Interleukin-2 (IL-2) and a platinum-based N-heterocyclic carbene (Pt-NHC) compound. Described in research published in Advanced Functional Materials (2023/2024), this nanoparticle-based system is intended to treat immunologically cold tumors, specifically pancreatic ductal adenocarcinoma (PDAC) and hepatocellular carcinoma (HCC) associated with advanced liver fibrosis. The Pt-NHC component acts as a type II immunogenic cell death (ICD) inducer, while IL-2 serves as a cytokine immunotherapeutic to activate effector T cells. The nanogel works by reducing the immunosuppressive phenotype of tumor-associated macrophages (TAMs), diminishing regulatory T cell (Treg) infiltration, and inducing type I interferon production. Preclinical studies have also explored its use in combination with losartan to further reduce desmoplasia and enhance the immunostimulatory state of the tumor microenvironment.
