IL4 Kinoid + IL13 Kinoid is an experimental conjugate vaccine combination designed to induce durable neutralizing antibody responses against the type 2 cytokines interleukin‑4 (IL‑4) and interleukin‑13 (IL‑13), key drivers of IgE production, eosinophilic inflammation, airway hyperresponsiveness, and mucus hypersecretion in allergic asthma. Each kinoid consists of a nonfunctional form of the cytokine covalently coupled to an immunogenic carrier protein (e.g., CRM197), converting the self‑cytokine into a T‑dependent antigen and eliciting endogenous polyclonal antibodies that neutralize IL‑4 and/or IL‑13 signaling through IL‑4Rα‑containing receptors.[1][5][6] In mouse and humanized mouse asthma models, dual vaccination with IL‑4 and IL‑13 kinoids significantly reduces serum IgE, airway eosinophilia, lung inflammation, mucus production, and bronchoconstriction, with effects lasting many weeks after immunization, supporting development as a long‑acting, cost‑effective alternative or complement to monoclonal antibodies targeting the IL‑4/IL‑13 pathway for allergic diseases.[1][2][5][6][9]
