**Ilixadencel + pembrolizumab** is an investigational combination therapy for cancer, utilizing **ilixadencel** (an allogeneic monocyte-derived dendritic cell medicinal product) administered intratumorally, together with **pembrolizumab** (a monoclonal antibody immune checkpoint inhibitor targeting PD-1)[1][5]. Ilixadencel is designed to prime the immune system by recruiting and activating endogenous immune cells, including natural killer (NK) cells and dendritic cells, thereby promoting an adaptive anticancer immune response. Pembrolizumab binds the PD-1 receptor on T lymphocytes, blocking its interaction with PD-L1/PD-L2 ligands, which allows T cells to attack cancer cells more effectively[2][4]. The rationale for combining these two modalities is to enhance T-cell activation and immune-mediated tumor destruction, potentially overcoming resistance to immune checkpoint inhibition. Developed primarily for advanced solid tumors such as head and neck squamous cell carcinoma, non-small cell lung cancer, and gastric/gastroesophageal junction adenocarcinoma, this combination is under clinical investigation but has not shown sustained efficacy in early clinical trials[1][5].
