IMA402 + checkpoint inhibitor is an investigational combination therapy developed by Immatics Biotechnologies. IMA402 is a next-generation, half-life extended, bispecific T-cell engaging receptor (TCER®) molecule designed to target a PRAME-derived peptide presented by HLA-A*02:01 on cancer cells. As an 'off-the-shelf' biologic, IMA402 works by redirecting and activating T cells to attack PRAME-expressing tumor cells, regardless of the T cells' intrinsic specificity. The combination therapy pairs IMA402 with an immune checkpoint inhibitor (ICI), such as pembrolizumab, to potentially enhance anti-tumor activity by overcoming immunosuppressive signals in the tumor microenvironment. This combination is being evaluated in Phase I/II clinical trials for patients with recurrent or refractory solid tumors, including cutaneous and uveal melanoma, ovarian cancer, lung cancer, and synovial sarcoma.
