imatinib + sorafenib

Imatinib + sorafenib is a combination therapy of two tyrosine kinase inhibitors with complementary mechanisms of action. This combination has been studied particularly in gastrointestinal stromal tumors (GIST) that have developed resistance to standard therapies, as well as in liver fibrosis models. Imatinib is a selective KIT tyrosine-kinase inhibitor that targets the BCR-ABL fusion protein, PDGFR, and c-KIT. It is considered standard first-line therapy for metastatic GIST and various leukemias[1][5]. Sorafenib is a multikinase inhibitor that targets RAF/MEK/ERK pathway and inhibits tumor angiogenesis by targeting VEGFR and PDGFR signaling in tumor vasculature[6]. When used in combination, these drugs have shown potential for stable disease in patients who have progressed on standard therapies, with one case study showing stable disease for nearly two years on the combination[1].
