Imatinib + toripalimab is a combination therapy being investigated for the treatment of advanced melanoma, specifically in patients harboring CKIT gene mutations. Toripalimab is a humanized IgG4 monoclonal antibody that targets the programmed cell death protein-1 (PD-1) receptor. By blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2), toripalimab acts as an immune checkpoint inhibitor, enhancing the body's anti-tumor immune response. Imatinib mesylate is a small molecule tyrosine kinase inhibitor (TKI) that targets several proteins, including BCR-ABL, platelet-derived growth factor receptor (PDGFR), and the proto-oncogene c-KIT (CD117). In the context of melanoma, imatinib is used to target the c-KIT signaling pathway, which is often dysregulated in specific subtypes of the disease. The combination is being studied by the Peking University Cancer Hospital & Institute to overcome the limited efficacy of PD-1 monotherapy in CKIT-mutant melanoma patients.
