Drug intelligence / Profile preview
Rytelo + azacitidine + venetoclax is an investigational three-drug combination regimen being evaluated for the treatment of relapsed or refractory acute myeloid leukemia (AML). The combination consists of imetelstat (Rytelo), a telomerase-inhibiting oligonucleotide developed by Geron; azacitidine (Vidaza), a hypomethylating nucleoside analog; and venetoclax (Venclexta), an oral selective BCL-2 inhibitor. This regimen is being evaluated in the Phase 1 IMAGINE trial (NCT07320235) to determine safety, tolerability, and preliminary efficacy, specifically focusing on finding the optimal biological dose of imetelstat in combination with azacitidine and venetoclax. Preclinical studies have demonstrated that the combination of imetelstat and venetoclax exhibits synergistic pro-apoptotic activity in AML cell lines and patient-derived samples.
