Overview

Imexon + gemcitabine is a combination of two small molecule chemotherapeutic agents investigated primarily for the treatment of advanced pancreatic adenocarcinoma. Imexon is an aziridine-derived iminopyrrolidone that acts as an inhibitor of ribonucleotide reductase, leading to cell cycle arrest in S phase and induction of apoptosis via mitochondrial dysfunction and oxidative stress. Gemcitabine is a nucleoside analog that also targets ribonucleotide reductase and inhibits DNA synthesis. The combination has shown synergistic cytotoxicity in preclinical models and was evaluated in phase I/II clinical trials for metastatic pancreatic cancer but did not demonstrate improved outcomes over standard therapy[1][3][4][6][8].
