Overview

IMGN151 + olaparib is a combination therapy under clinical investigation for the treatment of gynecologic cancers, particularly ovarian and other related cancers. IMGN151 is an antibody-drug conjugate (ADC) designed to deliver a cytotoxic payload specifically to cancer cells expressing certain surface antigens, thereby minimizing off-target toxicity. Olaparib is a small-molecule inhibitor of poly(ADP-ribose) polymerase (PARP) enzymes, which impairs DNA repair in BRCA-mutated or homologous recombination-deficient cancer cells, leading to cancer cell death. The combination is being evaluated to assess synergistic antineoplastic activity and improved outcomes in gynecologic oncology, likely leveraging IMGN151’s targeted cytotoxicity and olaparib’s DNA repair inhibition[1][3][5].
