Imlunestrant + abemaciclib is a **combination of imlunestrant**, a next-generation, oral, brain-penetrant selective estrogen receptor degrader (SERD) and pure estrogen receptor antagonist, and **abemaciclib**, a cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor. This combination is designed to deliver continuous estrogen receptor inhibition—including in cancers harboring *ESR1* mutations—while also blocking cell cycle progression via CDK4/6 inhibition. The combination demonstrated significantly improved progression-free survival compared to imlunestrant alone in patients with estrogen receptor (ER)-positive, HER2-negative advanced breast cancer that recurred or progressed following endocrine therapy. Safety was consistent with known profiles, with predominantly low-grade adverse effects.
