Imlunestrant + trastuzumab is an investigational combination therapy for the treatment of estrogen receptor–positive (ER+), human epidermal growth factor receptor 2–positive (HER2+) advanced breast cancer. Imlunestrant is a next-generation oral selective estrogen receptor degrader (SERD) that binds to and degrades the estrogen receptor, leading to inhibition of ER signaling—including in cancers with ESR1 mutations. Trastuzumab is a monoclonal antibody targeting HER2 and inhibits HER2-mediated signaling pathways. The combination aims to simultaneously block both ER and HER2 pathways in patients whose tumors express both targets. Early-phase clinical trials have shown that this combination is well tolerated with manageable side effects and demonstrates preliminary antitumor activity in patients with ER+/HER2+ advanced breast cancer who have received prior therapies[1].
