IMM-101 + anti-PD-1 is a combination immunotherapy consisting of IMM-101, a heat-killed whole cell preparation of Mycobacterium obuense that acts as a broad-spectrum immunomodulator, and an anti-PD-1 monoclonal antibody checkpoint inhibitor such as nivolumab or pembrolizumab. IMM-101 enhances innate and adaptive immune responses by promoting dendritic cell maturation and increasing antigen-specific CD8+ cytotoxic T lymphocyte and CD4+ Th1 cell responses. Anti-PD-1 monoclonal antibodies block the programmed cell death-1 (PD-1) receptor on T cells, preventing their inhibition by tumor PD-L1/PD-L2, thereby reinvigorating anti-tumor immune activity. Preclinical and clinical studies indicate that this combination may produce synergistic immune activation and anti-tumor efficacy, with observed encouraging responses in metastatic melanoma and ongoing evaluation in metastatic pancreatic cancer. The combination is under evaluation for durable response rates and acceptable safety in patients with advanced melanoma and pancreatic ductal adenocarcinoma[1][2][3][4].
