Overview

**immune checkpoint inhibitors + cetuximab** is a therapeutic combination used primarily in oncology, especially for certain advanced solid tumors. Immune checkpoint inhibitors (such as PD-1/PD-L1 or CTLA-4 inhibitors) act by blocking inhibitory signals on T-cells to restore and enhance anti-tumor immunity. Cetuximab is a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR), inhibiting cellular proliferation and survival signals and also mediating antibody-dependent cellular cytotoxicity. The rationale for this combination is based on the complementary mechanisms: cetuximab can modulate the tumor microenvironment and upregulate antigen presentation, potentially synergizing with immune checkpoint blockade to improve clinical response. This combination is under evaluation and sometimes clinical use in microsatellite stable (MSS) or proficient mismatch repair (pMMR) colorectal cancer and recurrent/metastatic head and neck squamous cell carcinoma (HNSCC), among other tumors, particularly after progression on first-line immunotherapy or chemotherapy. Preliminary studies suggest augmented anti-tumor activity, but the combination may confer an increased risk of immune-related adverse events (notably drug-induced lung injury). Large-scale randomized data are pending to confirm safety and efficacy[1][2][6][7].
