IMSA101 + immune checkpoint inhibitor is an experimental combination therapy involving **IMSA101**, a small molecule analog of cyclic GMP-AMP (cGAMP) and potent **STING** (stimulator of interferon genes) agonist, together with an **immune checkpoint inhibitor** (ICI), typically a PD-1 or PD-L1 inhibitor such as nivolumab or pembrolizumab. IMSA101 acts by activating the cGAS-STING pathway, which stimulates the innate immune system and enhances tumor detection and killing. Preclinical studies show that IMSA101 enhances antitumor activity both as monotherapy and in combination with ICIs, especially when delivered via intratumoral injection. The combination aims to trigger and amplify both innate and adaptive immune responses against tumors, potentially converting immunologically "cold" tumors to "hot," making them more responsive to checkpoint blockade. Current clinical trials are investigating this combination, particularly in patients with advanced solid tumors, oligometastatic non-small cell lung cancer, renal cell carcinoma, and oligoprogressive solid tumors[1][3][5].
