IMSA101 + immune checkpoint inhibitor is a combination therapy being developed by ImmuneSensor Therapeutics for the treatment of advanced solid tumors. IMSA101 is a small molecule agonist of the Stimulator of Interferon Genes (STING) pathway, designed as a cyclic dinucleotide analog of cGAMP. By activating the cGAS-STING pathway, IMSA101 stimulates the innate immune system, inducing the production of Type I interferons and other pro-inflammatory cytokines, and promoting the maturation of dendritic cells to enhance T-cell-mediated anti-tumor immunity. The addition of an immune checkpoint inhibitor (ICI), typically a PD-1 or PD-L1 antagonist, aims to overcome tumor-mediated immunosuppression and synergistically enhance the immune response, potentially turning 'cold' tumors into 'hot' tumors. This combination is currently being evaluated in Phase 2 clinical trials for various treatment-refractory malignancies, including head and neck, gastroesophageal, and breast cancers.
